The actress plays Nurse Nancy Corrigan on the show but has been noticeably absent from the latest season.
Nancy had moved away from Poplar after getting engaged to Roger Noble at the beginning of season 14.
In a new interview with Radio Times, Cusack confirmed she will be leaving the show.
The actress said she wanted to step out of her comfort zone after being part of the show for four years.
"Sometimes, you've got to step out of your comfort zone to grow.
"I've learnt so much, but I'm at the start of my career and I need to take a leap of faith," Cusack said.
Speaking about the cast of Call the Midwife, Cusack said she has made friends for life.
"It's bittersweet, really. I've made lifelong friendships there and I get to take those with me," she gushed.
The actress said she will also be upset leaving the show's crew behind.
"But when I think about not rocking up to set and seeing Tim, who's part of the grips team, with his guitar hanging out of his van and serenading people at half six in the morning… It really has been like a family."
The nurse is set to return in next Sunday's episode of Call the Midwife but the show's creators have not confirmed how Nancy's storyline will play out.
We're hoping the nurse finally gets her happy ending before she says goodbye to Poplar.
Call the Midwife continues next Sunday at 8 pm on BBC 1.
